The Stool Hanging Drop Test is a simple and rapid microscopic test used to discover motile bacteria in stool, for instance Vibrio cholerae, so it can cause cholera. The main function of the test is to help health care providers rapidly recognize the cause of watery diarrhea and provide early treatment in patients, especially during cholera outbreaks and areas endemic with cholera.
Process:
- A small amount of fresh stool is obtained and emulsified with saline on a glass slide.
- A coverslip is sealed and suspended over the cavity slide to create a hanging drop.
- The sample is then viewed through a microscope specifically designed to determine motility of bacteria.
When to get tested
If a patient has an acute onset of watery diarrhea, dehydration, or abdominal cramping, then this test is appropriate, particularly during a cholera outbreak, or after consuming unsafe water. This test is also used to confirm suspected bacterial GI infection.
Sample Type
Fresh stool sample (3-5 grams), stool obtained in a collection jar (sterile container is ideal).
Test Preparation
No fasting required, although tell your doctor about medications you are taking; though most medications have no impact on the outcome of the test.
Understanding Your Results
The presence of motile bacteria such as Vibrio cholerae supports the diagnosis of infection, and can assist in treatment management. Given that there are motile bacteria, but not Vibrio cholerae, further testing may be performed to exclude other causes of diarrhea or non-motile infection.
