The HLA-B27 Qualitative PCR Test is a blood test that looks for the HLA-B27 gene, which is associated with various autoimmune and inflammatory diseases such as ankylosing spondylitis, reactive arthritis, and some forms of eye inflammation. While having the gene does not mean that you will develop these diseases, it gives physicians further information to help with your symptoms and direct diagnosis and treatment.
Process
- Our technician will collect blood samples from a vein in your arm.
- The sample is sent to a laboratory where a DNA extract is prepared.
- The laboratory uses PCR (Polymerase Chain Reaction) to identify the presence of the HLA-B27 gene.
When to Get Tested
This test is appropriate if you have new unexplained joint pain, chronic back stiffness, eye inflammation, or if there is a family history of autoimmune diseases where your doctor is considering a disease related to HLA-B27, such as ankylosing spondylitis.

Test Preparation
For the test, there is an exception for fasting or special preparation. You can continue with eating and taking medications except when advised otherwise.
Understanding Your Results
A positive result indicates that the HLA-B27 gene has been detected and increases the risk of certain autoimmune disease. A negative result decreases the risk but does not eliminate the chance of disease. Your doctor will interpret the result considering other factors.
